So there's a lot of different factors that anda anda need to take into account. What do anda mean oleh "fighting?" Are anda asking based off the strongest the characters have ever gotten? (i.e. Madara Ten Tails Jinchurki atau Sasuke after obtaining power from Hagoromo). For the sake of this question, I'll base each one off of there own actual strength, up until they unlocked their mangekyou sharingan (not enternal).
If you're asking solely based off their taijutsu, I like to think that Madara is the strongest, with Itachi second, Saskue third and Shisui fourth.
In terms of raw strength, I think Madara would again come in first, Sasuke in second, Itachi in third, and Shisiu in fourth.
Now when anda get to fighting in general it gets a bit complicated, and its actual very hard to judge. Each one of them had there own unique and special abilities (I'm just gonna daftar some that I know are unique). Madara had the Izanagi, which when lets one change ones fate at the cost of losing berkata eye. Itachi had the Izanami, which cast a powerful genjutsu which affects the target through physical sensations shared between them and the user to perform the illusion, again this comes at the loss of the eye. Sasuke had the Amaterasu, the black flames, and Kagutsuchi, which helped him shape his Amaterasu, the use of these techniques gradually caused blindness. Shisiu had the Kotoamatsukami, an extremely powerful genjitsu which can be cast on the target without the need to make eye contact, this ability had decade long recharge rate.
Overall, I would say Madara is the best at fighting, with Itachi coming at a close detik because of his extreme skill and high intellect, Sasuke would be third, and Shisui fourth.
